清络通痹方对类风湿关节炎患者T淋巴细胞表达RANKL的影响(英文)

Effect of Qingluo-tongbi Granule on RANKL Expression of T Lymphocytes Extracted from RA Patients

目的研究清络通痹方(Qingluo-Tongbi Grannual,QLT)对类风湿关节炎(rheumatoid arthritis,RA)患者外周血T淋巴细胞表达RANKL(receptor activator of nuclear factor-κB ligand,破骨细胞分化因子)的影响。方法临床纳入15例RA患者,肘静脉抽血10ml,每个血样分为5组,包括正常对照组,阳性对照组,和QLT小、中、大三个剂量组。血样加药培养72小时后,加入FITC标记的CD3抗体和PE标记的RANKL抗体,采用流式细胞术检测RANKL的表达率。结果采用中剂量和大剂量QLT动物含药血清培养的T淋巴细胞RANKL表达率受到了显著的抑制,与正常对照组比较具有统计学差异(P<0.01)。结论 QLT可以有效下调RA患者外周血T淋巴细胞RANKL的表达。

Objective To study the effect of medicated serum containing Qingluo - tongbi Granule （QLT） on RANKL （ receptor activator of nuclear factor - KB ligand） expression of T lymphocytes extracted from peripheral blood of rheumatoid arthritis （RA） patients. Methods Blood samples extracted from veins of 15 RA cases were divided into 5 groups, including normal control, positive control, small - dose QLT, medium - dose as well as large - dose. Samples were cultured, processed under general program and tested with flow cytometer after antibodies of FITC labeled anti -human CD3 and PE labeled anti -human RANKL were added in. Results Compared with normal control group, RANKL expression of T lymphocytes treated with serum containing medium or large dose QLT was inhibited obviously in vitro （ P 〈 0.01 ）, and QLT demonstrate effect to suppress RANKL expression. Conclusion QLT is able to suppress RANKL over - expression of T lymphocytes of RA patients.
