摘要
目的检测Notch信号通路中相关基因在儿童急性T淋巴细胞白血病(T-ALL)中的表达水平,并探讨其在儿童T—ALL中的发病机制。方法采用实时定量RT—PCR方法检测23例初诊T—ALL患儿和20例正常对照儿童骨髓中Notch1、Notch2、Jagged1、Delta1的表达水平。结果儿童T-ALL组骨髓中受体Notch1的表达水平明显高于对照组,配体Jagged1的表达水平明显低于对照组,差异有统计学意义(P〈0.05);而受体Notch2、配体Delta1的表达在T-ALL组与对照组比较差异均无统计学意义(P〉0.05)。结论受体Notch1在儿童T—ALL中高表达,配体Jagged1在儿童T—ALL。中低表达,这与成人T—ALL中Notch相关基因的表达相一致,提示儿童T—ALL发病与Notch信号通路中基因的突变或者异常表达有关。
Objective To investigate the expression of Notch signaling pathway related genes in childhood T cell acute lymphoblastic leukemia (T-ALL) and investigate its role in the carcinogenesis ofchildhood T-ALL. Methods quantitative RT-PCR was used Real-time to detect the expression levels of Notch signaling cluding Notchl, Notch2, Jaggedl, genes including Notchl, Notch2, Jaggedl, Delta1 inthe bone marrow of 23 childhood T-ALL patients at the first diagnosis and 20 controls. Results The mean expression levels of Notch in 23 childhood T-ALL patients were statistically higher than those of 20 normal controls and the Jaggedl were lower than those of 20 normal controls (P 〈 0. 05 ). However, the mean expression levels of Notch2, Dehal had no significant difference with those of normal controls (P 〉 0. 05). Conclusions Notchl is over-expressed in childhood T-ALL while Jaggedl is moderately and weakly expressed in childhood T-ALL, suggesting that abnormal Notch signaling pathway is related with leukemogenesis of child T-ALL.
