摘要
ST细胞为一种广泛使用的兽用疫苗生产传代细胞,该研究利用一种新型的潮汐式生物反应器Bellocell进行了ST细胞的培养试验。通过3L转瓶培养扩增ST细胞,消化分散后以1.07×108个细胞接种对潮葡汐萄式糖反的应消器耗逐Be日llo增ce快ll?,为-5维00持,培细养胞7的天正,每常天生监长测,4细8h胞之数后量需、p要H每和天葡更萄换糖新值的的培变养化基。。试在验1期~间6天细的胞培养期间,观察到细胞每日增长倍数在1.2~2.4倍之间,接种后第6天细胞数量达到峰值3.17×109个,是接种之初的30倍。细胞数量达到109时,显微镜观察载体上的ST细胞,呈梭形、大小均一,台盼蓝染色结果证实细胞存活。试验成功实现了ST细胞的高密度培养,为以ST细胞为生产基质的兽用疫苗生产提供了一条简便快捷的途径。
ST cell was a continuous cell line which widely used in veterinary vaccine production. This study used a new type of tidal bioreactor, Bellocell, for the ST cell culture. ST cells were amplified by 3L roller bottle. When the cell number reached 1.07x l0s, dispersed cells were seeded into tidal reactor BellocellΘ-500, with 500 mL culture medium, and cultivated for 7days. In each day, the cell number, pH and glucose values change were monitored. When the cells number reached 109, the ST cells on the carrier showed spindle-shaped, uniform size in microscope, result of trypan blue staining confirmed that cells were surviving. This study successfully achieved a high density culture for the ST cell, that would provide a simple and efficient way for the production of veterinary vaccine with the ST cell.
