摘要
目的:探讨缺氧诱导因子1α(HIF-1α)和血管内皮生长因子(VEGF)在多发性骨髓瘤(MM)中的表达情况及两者的相关性和临床意义。方法:应用SP免疫组化法检测40例MM组和20例对照组患者骨髓组织中HIF-1α和VEGF的蛋白表达和分布。结果:MM组HIF-1α蛋白高表达(72.50%),显著高于对照组(35.00%),P<0.001;MM组VEGF蛋白表达率(67.50%),显著高于对照组(30.00%),P<0.05;HIF-1α在MM中的阳性表达与VEGF的高表达呈正相关(rs=0.529)。结论:HIF-1α和VEGF的表达与MM的发生发展密切相关且两者呈正相关,HIF-1α蛋白可能以转录激活的形式上调VEGF基因的表达,诱导血管生成,促进了MM的发生发展。
Objective: To explore the expression and significance of hypoxia-inducible factor-1α(HIF-1α) and vascular endothelial growth factor(VEGF) in multiple myeloma(MM).Methods: SP immunohistochemical staining technique was used to detect the expression of HIF-1α and VEGF in the myeloid tissue of 40 MM patients and 20 controls.Results: Both HIF-1α and VEGF were overexpressed in MM group as compared with the controls[(72.50% vs.35.00%;67.50% vs.30.00%,respectively);(P〈0.001;P〈0.05)],and HIF-1α was positively correlated with VEGF expression(rs=0.529).Conclusion: HIF-1α and VEGF expression are closely and positively correlated with the carcinogenesis and development of multiple myeloma,suggesting that HIF-1α protein may up-regulate the expression of VEGF gene by transcriptional activation that finally induces angiogenesis resulting in the genesis and development of this tumor.
