p57^(Kip2)在鼻咽癌中的表达及意义

摘要 目的揭示p57Kip2在鼻咽癌中的表达情况和临床意义,并分析其可能的机制。方法收集广州军区武汉总医院耳鼻喉科2005-06/2010-05月收治的鼻咽癌组织标本133例,对照组选取相同时间段该科室采集的鼻咽黏膜慢性炎症组织43例,实时荧光定量聚合酶链反应(real-time quantitative polymerase chain reaction,Realtime-PCR)法检测标本中p57Kip2以及人第10号染色体缺失的磷酸酶及张力蛋白同源基因(phosphatase and tension homolog deleted on chro-mosome ten,PTEN)的表达情况,分析p57Kip2在鼻咽癌中表达的意义及其与PTEN表达的相关性。结果 p57Kip2在鼻咽癌组织中的表达低于对照组织(P<0.01),p57Kip2水平与鼻咽癌转移呈显著负相关(P<0.01),但是与患者性别、年龄等因素无关(P>0.05),且p57Kip2在鼻咽癌中的表达与PTEN水平正相关。结论 p57Kip2可能在抑制鼻咽癌侵袭转移中发挥了重要作用,其机制可能是通过促进抑癌基因PTEN的表达。 Objective To reveal the expression and clinical significance as well as the possible mechanism of p57Kip2 in nasopharyngeal carcinoma. Methods A total of 133 cases with nasopharyngeal carcinoma were collected in Department of Otolaryngology in Wuhan General Hospital from June 2005 to May 2010, and 43 cases with chronic inflammation in na sopharyngeal mucosa were collected in the same period as control. The expression of p57Kip2 and phosphatase and tension homolog deleted on chromosome ten (PTEN) was detected by real-time quantitative polymerase chain reaction (Realtime- PCR). The significance of p57Kip2 and its correlations with PTEN expression were analysed. Results The expression of p57Ripe in nasopharyngeal carcinoma were significantly lower than that in the control (P〈0. 01 ). p57Kip2 levels was negatively correlated to nasopharyngeal carcinoma metastasis (P〈0. 01), but not significantly correlated to gender, age or other factors (P〉0. 05). p57Kip2 was positive correlated to PTEN in nasopharyngeal carcinoma. Conclusion p57Kip2 may be important in the inhibition of nasopharyngeal carcinoma invasion and metastasis by promoting the expression of tumor suppressor gene PTEN. The underlying mechanism warrants further investigations.
