摘要 死亡相关蛋白激酶(DAPK)是一种新的钙调蛋白(CaM)调节的丝/苏氨酸激酶,是凋亡的正性调节因子。细胞凋亡被认为是控制和治疗肿瘤的最有效方法之一,它与肿瘤的发生、发展和转移有着密切的联系。而DAPK参与多条途径诱导的细胞凋亡,被公认为是一种肿瘤抑制基因。在此我们将重点讨论DAPK促进细胞凋亡的机制,为靶向治疗肿瘤提供方向和理论依据。 Death-associated protein kinase(DAPK) is the founding member of a newly classified family of calmodulin(CaM)-regulated Ser/Thr kinases that functions as a positive mediator of apoptosis.Apoptosis plays an important role in tumor initiation,pro-gression and metastasis,whose regulation may be an effective way for tumor therapeutics.DAPK is considered as an tumor suppressor involved in diverse apoptosis pathways.This review summarizes relevant knowledge of DAPK family with an emphasis on potential mechanisms of DAPK family-mediated apoptosis and its potential to serve as a novel therapeutic target in oncology.
