摘要
目的利用基因芯片技术探讨附子"温阳补火"温热效应的产生机制。方法 SPF级Wistar雄性大鼠分为两组,分别灌胃附子水煎剂和生理盐水。给药20 d后,提取肝脏总RNA,应用Illumina大鼠全基因组表达谱基因芯片研究其"温阳补火"功效的分子机制。结果附子组与对照组比较有592条基因差异表达,催化活性基因功能为较显著性基因功能,42条涉及氧化还原酶活性(上调26条,下调16条),其中7条与氧自由基的生成和清除有关,3条涉及性激素代谢过程。结论附子可下调超氧阴离子生成催化酶基因水平,上调自由基清除相关基因表达水平,减少自由基生成,促进其清除;调控性激素代谢相关基因表达,促进性激素转化,减少灭活,发挥一定抗氧化、抗衰老作用,这可能是其温热效应机制之一。
Objective To investigate the molecular mechanism of warming yang and reinforcing fire of aconite root by gene chip technology. Methods Twenty male SPF Wistar rats were randomly divided into two groups, respectively administrated intragastrically with aconite root decoction and normal saline. 20 days later, the rat liver gene expressions in each group were analyzed by whole genome microar- ray made by Illumina Company to probe into the molecular mechanism of warming yang and reinforcing fire function of aconiti root. Results 592 strips of gene differential expressions were found between the two groups, catalytic activity being a significant gene function,42 strips (up-regulation 26; down-regulation 16) were related to dehydrogenase activity. Seven of the 42 strips were involved the generation and re- moval of oxygen radical. Three were related to the metabolic process of sex hormone. Conclusions Aconite root may inhibit the expressions of genes related to catalytic activity of superoxide anion generation, increase the expressions of genes related to radicals scavenging , reduce the generation of free radicals and promote scavenging. In addition, it may regulate the expressions of genes related to the metabolism of sex hormones, facilitate the conversion of sex hormones, reduce their inactivation , which maybe the one molecular mechanism of warming yang and reinforcing fire of aconite root.
