摘要
目的观察酸枣仁汤组分配方对高架十字迷宫大鼠海马单胺类递质及其代谢物的影响,探讨其抗焦虑效用的有效组分及作用机理。方法采用国际上通用的高架十字迷宫焦虑动物模型,观察酸枣仁汤组分配方对大鼠行为学的影响,高效液相色谱法测定大鼠海马单胺类递质去甲肾上腺素(NE)、5-羟色胺(5-HT)、多巴胺(DA)及其代谢物的含量。结果与空白对照组比较,全组分配方组和有效组分配方组及地西泮组大鼠进入开放臂次数比、在开放臂停留时间比均显著升高(P<0.01或P<0.05);有效组分配方组和地西泮组大鼠海马5-HT的含量均见显著降低(P<0.05),全组分配方组海马5-HT含量呈降低作用趋势;有效组分配方组大鼠海马高香草酸(HVA)含量显著升高(P<0.01)。结论酸枣仁汤全组分配方及其有效组分配方与地西泮具有某些相同的抗焦虑作用机制,可能涉及到降低海马5-HT含量;全组分配方及其有效组分配方对大鼠海马单胺类递质代谢的影响有所不同。
Objective To observe the influence of component formula of Suanzaoren Tang on hippocampus monoamine neurotransmitters and their metabolites in rats in elevated plus maze test ( EPM), and discuss its effective components and mechanism of anti-anxiety. Methods The influence of component formula of Suanzaoren Tang on rat ethology was observed by using international common-used anxiety animal model in EPM. The levels of monoamine n including noradrenaline ( NE), 5-hydroxytryptamine (5-HT) and dopamine (DA) and their metabolites Were detected by applying HPLC. Results Compared with blank group, the ratio of entering times and retention time ratio in open arm increased significantly in whole component formula group (whole CF group), effective component formula group (effective CF group) and diazepam group (P 〈0. O1 or P 〈0.05). The level of 5-HT decreased significantly in effective CF group and diazepam group, and showed a decreasing trend in whole CF group. The level of hippocampus homovanillic acid (HVA) increased significantly in effective CF group (P 〈 O. O1 ). Conclmion The whole component formula and effective component formula of Suanzaoren Tang are similar to diazepam in anti-anxiety mechanism to some extent, which may involve the decrease of hippoeampus 5-HT level. The whole component formula and effective component formula have different influences on the metabolism of hippocampus monoamine neurotransmitters in rats.
