PDGF-BB及其受体PDGFR-β在人大肠癌组织中的表达及与CAFs的关系 被引量:3

Expression of PDGF-BB and PDGFR-β and its relations to cancer-associated fibroblasts in colorectal cancer

摘要 目的:检测PDGF-BB及其受体PDGFR-β在大肠癌组织中的表达,分析其与癌相关成纤维细胞(CAFs)的关系,探讨两者在肿瘤发生发展中的作用。方法:采用免疫组织化学Elivision二步法,检测106例大肠癌组织及20例癌旁正常对照组织中PDGF-BB及其受体PDGFR-β的表达,用α-SMA标记CAFs,分析PDGF-BB、PDGFR-β的表达情况及与临床病理参数、CAFs之间的关系。结果:PDGF-BB及受体PDGFR-β在癌组织中阳性表达显著高于癌旁正常大肠组织[PDGF-BB:58.49%(62/106)vs 10.00%(2/20);PDGFR-β:78.30%(83/106)vs25.00%(5/20),P均<0.05];PDGF-BB在大肠癌癌细胞中的表达与间质中PDGFR-β的表达呈显著正相关(r2=0.4254,P=0.033)。PDGF-BB、PDGFR-β表达水平与患者TNM分期、淋巴结转移有关,而与患者的年龄、肿瘤大小、组织类型、分化程度均无关。106例大肠癌组织标本间质中α-SMA阳性表达率为64.15%(68/106),癌旁正常肠组织的阳性表达率为20.00%(4/20),两组比较差异有统计学意义(P<0.05);且α-SMA阳性表达与患者淋巴结转移呈正相关(r=0.095,P=0.030)。PDGFR-β在人大肠癌组织标本中的表达还与CAFs正相关(r=0.227,P=0.019)。结论:PDGF-BB及其受体PDGFR-β在大肠癌组织中的表达与CAFs相关,其相互作用可能参与了大肠癌的发生发展及淋巴结转移。 Objective: To evaluate the relationship between the expression of PDGF-BB,PDGFR-β and cancer-associated fibroblasts(CAFs) in colorectal cancer.Methods: Expressions of PDGF-BB and PDGFR-β were detected using the immunohistochemical Elivision method,in 106 colorectal cancer and 20 adjacent normal tissues.CAFs were labeled by α-SMA.The relationship between PDGF-BB,PDGFR-β and CAFs or the clinicopathological features of colorectal cancer were then analyzed.Results: The expressions of PDGF-BB and PDGFR-β in colorectal cancer were both higher than that of adjacent normal tissues [PDGF-BB:58.49%(62/106)vs10.00%(2/20);PDGFR-β:78.30%(83/106)vs 25.00%(5/20),both P〈0.05].The expression of PDGF-BB in colorectal cancer was significant correlated with the expression of PDGFR-β(r2=0.4254,P=0.033).Expression of PDGF-BB and PDGFR-β were positively correlated with TNM stage and lymph nodes metastasis,but no relationships with sex,age,histology type and,cell differentiation were found.α-SMA was also over-expressed(64.15%) in colorectal cancer tissues,while its expression was(20.00%) in the normal tissues(P〈0.05).Expression of α-SMA was positively correlated with lymph nodes metastases(r=0.095,P=0.030) and PDGFR-βexpression(r=0.227,P=0.019).Conclusion: In colorectal cancer tissues,expressions of PDGF-BB and PDGFR-β are closely related with CAFs.The interaction among them might be involed in the development of colorectal cancer.
