S-腺苷蛋氨酸对大鼠肝缺血再灌注损伤后白蛋白和前清蛋白的影响 被引量:4

摘要 目的:观察S-腺苷蛋氨酸(思美泰)对SD大鼠肝缺血再灌注损伤后白蛋白和前清蛋白的影响。方法:将72只SD大鼠随机分为3组:假手术组(SHAM组)、缺血再灌注对照组(IR组)、缺血再灌注腺苷蛋氨酸干预组(SAM组),每组24只。1)SHAM组打开腹腔,游离肝十二指肠韧带,不阻断肝脏血供,20 min后再关腹。2)IR组和SAM组开腹后游离肝十二指肠韧带,用无创微血管夹夹闭肝蒂,持续20min后松开血管夹然后关腹。Sham组、IR组术后均予生理盐水1 mL(/kg.d)腹腔注射,SAM组予思美泰100 mL(/kg.d)腹腔注射。3)3组分别于术后第1、4、7、10天处死6只大鼠,测定血清前清蛋白和白蛋白的水平变化,同时取肝左叶部分组织行电镜观察。结果:IR组与SAM组血清白蛋白术后第1、4、7、10无差别(P>0.05);IR组与SAM组之间血清前清蛋白水平比较,术后第1天无差别(P>0.05),而术后第4、7、10天SAM组的血清前清蛋白水平高于IR组的水平(P<0.01)。电镜显示术后第4、7、10天SAM组肝细胞内质网和线粒体数量明显多于明显于IR组。结论:腺苷蛋氨酸治疗能够提高血清前清蛋白水平,减轻肝细胞病理损害。 Objective: To investigate the influence of S-ademetionine on serum level of pre-albumin and albumin in rat liver after ischemia-reperfusion injury.Methods: The total 72 healthy males sprague-Dawle rats were randomly divided into three group:sham operation group(SHAM group),ischemic-reperfusion group(IR group),S-ademetionine group(SAM group),24 rats per group.Rats in sham group were administered with the opening and suture of the abdomen with equal NS by intraperitoneal injection.Rats in ischemic-reperfusion group underwent ischemia reperfusion with equal NS by intraperitoneal injection,while Rats in S-ademetionine group was performed with both 10% S-ademetionine and ischemia-reperfusion.All rat ischemia-reperfusion models were established by Pringle's method.1 hour after 20-minute schemic-reperfusion,intraperitoneal injection was given with the dosage of 1 ml /(kg·d) NS or 100 mg /(kg·d) S-ademetionine.Blood samples were collected and analyzed for the levels of serum albumin and prealbumin and hepatic tissue changes of the ultro-micro structure were observed at the 1st,4th,7th,10th day after 20-minute ischemia-reperfusion.Results: Comparing the level of serum albumin in SAM group and IR group,it was at the same level at the first,fourth,seventh,tenth day(P〈0.05),while the level of serum albumin in SAM group and IR group,it was at the same level at the first day(P〈0.05),different from each other at fourth,seventh,tenth day(P〈0.01).The damage of endoplasmic reticulum and mitochondrial in SAM Group had been lighter than in the IR group and the number of endoplasmic reticulum and mitochondria in SAM group higher than in IR group at fourth,seventh,tenth day.Conclusion: S-ademethionine can increase serum prealbumin level,relieve the pathological damage of hepatic cell.
