p53选择性剪接异构体在胃癌中表达的研究 被引量:2

摘要 目的探讨p53的5种选择性剪接异构体p53β、p53γ、Δ133p53、Δ133p53β和Δ133p53γmRNA在胃癌和癌旁组织中的表达情况及其与胃癌发生、发展的关系。方法收集2010年3月至2010年6月15例胃癌根治术患者的手术切除标本,采用巢式RT-PCR检测胃癌组织与癌旁组织中p53β、p53γ、Δ133p53、Δ133p53β和Δ133p53γ共5种异构体mRNA的表达情况,并进行统计学分析。结果在15例胃癌组织与癌旁组织中均未检测出p53γ、Δ133p53β和Δ133p53γmRNA的表达。Δ133p53在胃癌组织中的表达率为73.3%(11/15),在癌旁组织中为20.0%(3/15),差异有统计学意义(P=0.009)。p53β在胃癌组织中的表达率为20.0%(3/15),在癌旁组织中为66.7%(10/15),差异有统计学意义(P=0.0253)。结论Δ133p53 mRNA和p53βmRNA在胃癌和癌旁组织中的表达情况不同,这两种异构体的差异性表达与胃癌的发生发展有关。 Objective To investigate the mRNA expression of p53 splice variants-p53β,p53γ,Δ133p53,Δ133p53β and Δ133p53γ in gastric carcinoma tissues and adjacent non-cancerous tissues,and explore their roles in tumor genesis,development and progression of gastric carcinoma.Methods Fifteen pairs of fresh gastric carcinoma tissues and their adjacent non-cancerous tissues were obtained from radical gastrectomy in the Affiliated Hospital of Weifang Medical College from March 2010 and June 2010.All the samples were examined for mRNA expression levels of p53 splice variants: p53β,p53γ,Δ133p53,Δ133p53β and Δ133p53γ by nested RT-PCR.Statistical analysis was performed according to experimental data.Results p53γ,Δ133p53β and Δ133p53γ were not detected in both carcinoma tissues and adjacent non-cancerous tissues.Δ133p53 was detected in 11 out of 15 carcinoma tissues(73.3%) and in 3 out of 15 adjacent non-cancerous tissues(20.0%) with statistic significance(P=0.009).p53β was detected in 3 out of 15 gastric carcinoma tissues(20.0%) and in 10 out of 15 adjacent non-cancerous tissues(66.7%) with statistic significance(P=0.0253).Conclusion The expression of Δ133p53 mRNA and p53β mRNA were different in gastric carcinoma.The different expression of the p53 isoforms may be involved in growth,proliferation and metastasis of gastric carcinoma.
