细胞色素P450酶及儿茶酚-O-甲基转移酶蛋白在子宫内膜癌中表达半定量研究 被引量:1

Expression of CYP450 1B1 and COMT protein in human endometrial cancer

摘要 目的探讨细胞色素P450酶(cytochromeP4501B1,CYP1B1)及儿茶酚-O-甲基转移酶(catechol-O-methytransferase,COMT)在子宫内膜癌发病机制中的作用。方法采用Western Blot技术检测2006-2008年河北医科大学第四医院子宫内膜癌组织39例(病例组)及正常子宫内膜组织22例(对照组)中CYP1B1及COMT蛋白的表达,并对两者进行比较。结果病例组中CYP1B1蛋白的表达量(3.637±1.697)明显高于对照组(2.371±1.145)(P=0.003);而S-COMT、M-COMT在病例组中的表达量(分别为1.104±0.576、0.970±0.461)明显低于对照组(分别为1.602±0.665,2.091±0.609)(P=0.001,P=0.000)。病例组CYP1B1/S-COMT及CYP1B1/M-COMT明显高于对照组(P=0.002,P=0.000)。三期子宫内膜癌中,CYP1B1及S-COMT的差异有统计学意义(P=0.000,P=0.001)。结论 CYP1B1与子宫内膜癌的发生发展有密切关系,可能成为子宫内膜癌治疗的新靶点及预后的指标。COMT在子宫内膜癌的发展中起到了保护作用。 Objective To explore the effect of CYP1B1 (cytochromeP450 1B1) and COMT (catechol-O-methytrans-ferase) protein in the etiology of endometrial cancer. Methods Western blot was used to detect the expression of CYP1B1, COMT protein in 39 endometrial cancer and 22 normal endometrial tissues, which were collected during 2006- 2008 in the Fourth Hospital of Hebei Medical University, then to compare the difference. Results The relative expression of CYP1B1 in the case group (3. 637± 1. 697) was significantly higher than that in the control group (2. 371±1. 145 ) ( P = 0. 003 ) ; while the S-COMT, M-COMT expression in the case group ( 1. 104±0. 576,0. 970±0. 461 ) was significantly lower than that in the control group ( 1. 602±0. 665,2. 091 ±0. 609 ) ( P = 0. 001, P = 0. 000 ). The CYP1B1/S-COMT and CYP1BI/M-COMT in the endometrial cancer patients were significantly higher than controls(P = 0. 002,P =0. 000). The expression of CYP1B1 and S-COMT in three stages were statistically different (P = 0. 000,P = 0. 001 ). Conclusion CYP1B1 has a close association with the occurrence and development of endometial cancer, maybe a novel target for endometrial cancer therapy. COMT plays a protective role in the occurrence of endometrial cancer.
