慢病毒-绿色荧光蛋白载体转染人眼睑成纤维细胞的研究

摘要 目的将慢病毒-绿色荧光蛋白载体感染人眼睑成纤维细胞,为诱导性多潜能干细胞研究奠定基础。方法采用0.25%dispeasⅡ酶冷消化联合组织块贴壁法培养人眼睑成纤维细胞,免疫组化SP法鉴定细胞种类,将慢病毒-绿色荧光蛋白载体以不同的感染复数(MOI值)感染细胞,并于感染48 h后流式细胞仪检测感染效率。结果该实验方法能获得大量的成纤维细胞,并且纯度高,当MOI值为40时,其感染效率为95.37%。结论慢病毒-绿色荧光蛋白载体能够高效稳定感染人眼睑成纤维细胞表达绿色荧光蛋白,是诱导性多潜能干细胞技术研究的理想种子细胞。
