慢盆消炎方对慢性子宫内膜炎模型大鼠MMP-9、iNOS的影响 被引量:5

摘要 目的:观察慢盆消炎方对慢性子宫内膜炎模型大鼠血液流变学及子宫内膜组织基质金属蛋白酶-9(MMP-9)、诱导性一氧化氮合酶(iNOS)表达的影响。方法:雌性SD大鼠随机分为七组,每组10只。A组为正常对照组,B组为模型对照组,C组为妇乐冲剂组,D组为多西环素组,E组、F组、G组分别为慢盆消炎方低、中、高剂量组。采用阴道接种支原体+子宫注入混合菌株法制作慢性子宫内膜炎大鼠模型。造模后,各组给予相应的药物,连续灌胃给药20 d。给药结束后采血进行血液流变学检测,取大鼠子宫进行组织形态学观察,免疫组化法检测大鼠子宫内膜MMP-9、iNOS的表达情况。结果:模型对照组子宫内膜MMP-9、iNOS蛋白的表达明显高于A组(P<0.05);各用药组均能不同程度地减少MMP-9、iNOS表达;且慢盆消炎方中、高剂量组效果优于妇乐冲剂组、多西环素组。差异具有统计学意义(P<0.05)。结论:慢盆消炎方能改善慢性子宫内膜炎模型大鼠子宫内膜组织结构,减少炎性细胞浸润;可以有效改善慢性子宫内膜炎模型大鼠血液流动性;并能不同程度抑制炎症因子MMP-9、iNOS的表达。 Objective: To observe the effect of Manpen Xiaoyan Prescription on blood rheology and MMP - 9, iNOS of model rats with chronic endometritis. Methods: The female SD rats were randomly divided into seven groups, I0 in each group. Group A was the normal control group, group B model control group, group C Fule Granules group, group D doxycycline group, while group E, group F and group G were respectively low, medium and high dose of Manpen Xiaoyan Prescription. Model rats with chronic endometritis were produced by using vaginal inoculation of mycoplasma and uterus infusion of mixed strains. After the model building, each group was given corresponding drugs, continuous intragastric administration for 20d. The hemorheological detection was made and rat uterus tissue morphology was observed after the administration drug, the endometrial MMP -9, iNOS expression was detected with immunohistochemistry. Results: The endometrial MMP -9 and iNOS protein expression in the mod- el control group was significantly higher than that of group A ( P 〈 0.05 ) ; MMP - 9, iNOS expression was decreased in each drug group ; meanwhile, the effects of medium and high dose group of Manpen Xiaoyan Prescription were better than those of Fule Granules group and doxycycline group. The difference was statistically significant (P 〈 O. 05 ). Conclusion : Manpen Xiaoyan Prescription can improve the tissue structure of chronic endometritis model rat, reduce the infiltration of inflammatory cells; can effectively improve the blood flow of model rat ; and can inhibit the inflammatory factor MMP -9, iNOS expression in different degrees.
