LKB1蛋白在胃癌组织中的表达及其临床意义

Expression of LKB1 in gastric cancer and its clinical significance

目的探讨LKB1蛋白在胃癌组织中表达的临床意义,为胃癌的诊断及预后判断提供新指标。方法采用免疫组化过氧化物酶标记链霉卵白素法(SP法)检测胃癌组织中LKB1蛋白的表达水平,并与患者的年龄、性别、肿瘤大小、浸润深度、分化程度、淋巴结转移和远处转移、生存时间等临床病理因素进行相关性分析研究。结果 LKB1蛋白在正常胃上皮组织、胃癌组织及转移淋巴组织中均有表达,3组之间阳性表达率差异有统计学意义(P<0.01)。LKB1蛋白表达与患者性别、年龄、肿瘤大小、部位、分化程度、浸润深度均无相关关系;而与淋巴结转移、远处转移及临床分期(TNM)相关(P<0.05)。LKB1蛋白阴性组预后与阳性组比较差异有统计学意义(P<0.05)。结论 LKB1基因蛋白表达下降与胃癌患者发生淋巴结转移和远处转移相关;测定胃癌组织中的LKB1蛋白的表达,有助于判断患者的预后。

Objective To investigate the clinical significance of LKB1 expression in gastric carcinoma, in order to provide a new target for the clinical diagnosis and prognosis. Methods The expression levels of LKB1 protein in the organization were tested by immunohistochemistry of peroxidase labeled streptavidin egg sensitized （SP）, and the aggregated analysis combining the results with the clinical pathological factor such as age, gender, tumor size, depth of invasion, differentiation, lymph node metastasis and distant metastasis and so forth were made. The further study of the relationship between the expression of LKB1 protein and the survival time of patients was also carried out. Results LKB1 protein was expressed in normal gastric epithelial tissue, gastric cancer and the metastatic cancer in lymphoid tissue. The positive rate of them have a significant difference. There was no significant difference between the expression of LKB1 protein and patient gender, age, tumor size, location, degree of differentiation and the depth of invasion. However, there was significant difference in lymph node metastasis, distant metastasis and TNM stage （P 〈 0.05）. The prognosis of the negative group of LKB1 protein compare of the positive group, the difference is statistically significant （P 〈 0.05）. Conclusions LKB1 gene is related to the occurrence and development of gastric cancer. There is correlation between the down-regulation of the expression of LKB1 gene and occurrence of lymph node and distance metastasis in patients with gastric cancer. Determination of gastric cancer in the expression level of LKB 1 protein will conduct to decision the prognosis.
