摘要
目的:探讨CD133+细胞的生物学特性,分析CD133作为胶质瘤干细胞标记的可行性。方法:利用免疫荧光染色检测U251细胞中CD133、巢蛋白(nestin)、神经元特异性烯醇化酶(NSE)、神经胶质细胞纤维酸性蛋白(GFAP)等蛋白表达,分析CD133+细胞与CD133-细胞中巢蛋白、NSE、GFAP阳性细胞存在的差异。结果:U251中CD133+细胞比例为0.060±0.003,在CD133+细胞中,巢蛋白阳性细胞,NSE和GFAP阴性细胞比CD133-中明显增多;在CD133-细胞中,也存在巢蛋白阳性细胞,数目较CD133+中明显减少,相反巢蛋白阴性细胞,NSE和GFAP阳性细胞明显增多。结论:胶质瘤U251细胞存在CD133+细胞,在CD133阳性和阴性细胞中,均存在干细胞样细胞,但是在CD133+细胞中此类细胞更多。
Objective: To explore the biological characteristics of CD133 + cells and analyze the feasibility of CD133 as the marker in glioma stem cells.Methods: The expression of CD133,nestin,neuron-specific enolase(NSE),glial fibrillary acidic protein(GFAP) were detected by immunofluorescence in U251 glioma cells.The differences of the nestin,NSE,GFAP-positive cells were analyzed between CD133 + cells and CD133-cells.Results: In the U251 cells,the CD133+ cells composed to 0.060±0.003.In CD133+ cells,nestin-positive cells,NSE and GFAP-negative cells increased significantly;in CD133-cells,on the contrary,nestin negative cells,NSE and GFAP positive cells increased significantly.Conclusion: The CD133+ cells exist in U251 glioma cells,which have stem cell characteristics.
