摘要
以自行合成的叠氮标记的甘露糖进行细胞代谢,代谢后以叠氮化的唾液酸替代天然的唾液酸于细胞糖蛋白的糖链中呈现,依赖于点击化学方法,建立分离唾液酸化糖蛋白的方法,并研究gp96蛋白在不同癌细胞系中的唾液酸化程度.经酶联免疫吸附反应检测各细胞系均含有gp96蛋白,且含量无显著差异.而分析gp96的生物素化和唾液酸修饰程度,发现gp96在不同癌细胞中存在唾液酸修饰上的差异,在胰腺腺癌细胞中,gp96的唾液酸化程度的显著降低,可能直接影响其与免疫系统的相互作用.
Mannose with azide group was synthesized and metabolized by cells and presented on glycoproteins.Depending click chemistry method,a strategy to enrich sialylated glycoprotein was established,and the level of sialylation on gp96 in different cancer cells was investigated.After analyzing by ELISA,it was found that gp96 expressed in the three cell lines were no significant difference.However,the level of biotinylation and sialylation of gp96 was different.The results showed that sialylation on gp96 in SW1990 was significant lower than Hela and A549,and this suggested different immunological and pathological properties in pancreatic adenocarcinoma cells.
