摘要
目的探讨木犀草素是否可以对抗高脂饮食介导的血管功能损害。方法 30只C57BL/6J小鼠分正常饮食对照组(10只),高脂膳食组(对照组,10只)和高脂膳食加2%木犀草素组(木犀草素组,10只),干预16周,观察:①血压水平的变化;②胸主动脉血管功能的变化及长期木犀草素预防性饮食干预对血管功能的影响。③木犀草素短期孵育对肥胖小鼠血管功能的影响及机制。结果①小鼠在给予高脂膳食16周后,血压水平显著高于正常饮食对照组及高脂饮食+木犀草素组,P<0.05。②长期高脂膳食损害小鼠胸主动脉的血管内皮依赖性舒张功能,P<0.05;而木犀草素预防性膳食干预可防止血管内皮依赖性舒张功能的损害。③木犀草素孵育可改善高脂小鼠胸主动脉的内皮依赖性舒张功能,但该作用可被PPARγ的特异性阻断剂GW9662阻断。结论木犀草素可预防高脂膳食介导的小鼠胸主动脉血管内皮功能损害,该作用可能与激活PPARγ有关。
Object To investigate whether the Luteolin can prevents the impaired of endothelium dependent vasorelaxation induced by high fat diet. Methods 30 mice were divided into three groups, the normal chow group (10 mice), the high fat diet group (]0 mice), the high fat diet+2%Luteolin group, the intervention time is 16 weeks. We observe the blood pressure variation, the changes of the vascular function of the mice aorta and the effect of dietary Luteolin to the changes of the vascular fimction; we also study the effects of Luteolin incubation to the aorta flmction of the obesity mice. Results We found that after 16 weeks of high fat diet administration, the blood pressure was increased, P〈 0.05 versus control and HD+Luotelin groups. Long time high fat diet lead to the impaired of endothelium dependent vasorelaxation of mice aorta, dietary Luotelin prevents the impaired of endothelium dependent vasorelaxation of mice aorta. We also found that Luotelin incubating can improved the endothelium dependent vasorelaxation of obesity mice aorta, but this effect maybe inhibited by the PPAR'y antagonist GW9662. Conclusion Luotelin can prevents the impaired of endothelium dependent vasorelaxation of mice aorta induced by high fat diet, this effect may be due to the activation of PPART.
