双标cajal样间质细胞及一氧化氮合酶-1细胞在人膀胱组织的分布

摘要 目的探讨cajal样间质细胞(ICCs)及一氧化氮合酶-1(NOS-1)神经元细胞在人膀胱组织中的分布规律及意义。方法人膀胱全层组织冰冻切片,CD117免疫荧光结合NOS-1免疫荧光双重标记染色。结果激光共聚焦显微镜下观察CD117表达阳性的ICCs及NOS-1阳性神经元细胞广泛分布于人膀胱组织内,发现ICCs可表达NOS-1,总体上双重染色未发现两种细胞有明显的共表达,但可见NOS-1表达阳性神经元细胞的附近存在较多ICCs。结论人膀胱中的ICCs可能是膀胱自主节律性运动的起搏细胞,NOS-1阳性神经元细胞可能以ICCs作为靶点共同参与对膀胱舒缩活动的综控。 [Objective] To investigate the cajal-like interstitial cells (ICCs) and neuronal nitric oxide synthase (NOS-1) neurons cells in human bladder tissue's distribution and significance. [Methods] Using full-thickness frozen sections for human bladder tissue, CD117 with NOS-1 immunofluoreseenee are double labeling immunofluo-rescenee staining. [Results] Under eonfocal laser scanning microscope, CD117 positive of ICCs and NOS-l-positire neurons are widely distributing in the human bladder tissue, finding that ICCs could express NOS-1. Overall, double staining on two types of cells have not been found a clear co-expression, but the expression of NOS-l-positire neurons in the vicinity of which we can see lots of ICCs. [ Conclusion] The ICCs in human bladder may be the pacemaker cells which have independent rhythmic movement and the NOS-1 positive neurons may participate in ICCs as a target for bladder contractile activity of the integrated control.
