摘要 目的通过对内脏高敏感大鼠血清、结肠促肾上腺皮质激素释放激素(CRF)表达的研究,探讨外周CR F在内脏高敏感中的作用。方法取出生后8~21 d的Wistar大鼠,实验组每天直肠注射0.6%的冰醋酸0.3~0.5 mL,对照组给予同样剂量的生理盐水。8周龄时行直肠扩张评估内脏敏感性,记录腹部回缩反射(AWR)为3分时的注水量。心脏穿刺取血,ELISA法检测大鼠血清CR F蛋白含量,取降结肠组织,行CR F免疫组化。结果与对照组相比,实验组大鼠AWR为3分时的注水量明显减少(P〈0.05);CR F阳性面积在实验组大鼠结肠中较对照组明显增强(P〈0.01);CRF强度值在实验组大鼠结肠中较对照组明显增强(P〈0.05);CRF血清含量在实验组较对照组明显增强(P〈0.01)。结论血清、结肠CRF增加可能是内脏高敏感的发病机制之一。 [ Objective ] Through research of the expression of corticotropin releasing factor (CRF) in colon and plasma of rat with visceral hypersensitivity, to investigate the role of CRF in the pathophysiology of visceral hypersensitivity. [Methods] 8 to 21 days after birth,the Wistar rats of the experimental group were injected with daily recta glacial acetic acid 0.6% 0.3N0.5 mL, the control group were injected the same dose of saline. The visceral sensitivity of 8-week-old rats was evaluated by colorectal distention, and recorded the water injection rate when the score of abdominal withdrawal reflex (AWR) was 3. The plasma was collected to detect the level of plasma CRF by ELISA. The distal descending colon was taken to detect for CRF immunohistochemisty. [Results] Compared with the control group, the water injection rate of the experimental group showed a significant decrease when the score of AWR was 3 (P 〈0.05); CRF positive area in colon in the experimental group significantly increased (P 〈0.01); CRF positive intensity in colon in the experimental group significantly increased (P 〈0.05); CRF in plasma in the experimental group significantly increased (P 〈0.01). [ Conclusion] The increase of CRF in plasma and colon of rat with visceral hypersensitivity may be a possible cause of visceral hypersensitivity.
