摘要
目的初步研究ERβ在青春期脊柱线性生长中的调控作用。方法取相当于人青春前期的5周龄昆明小鼠36只,其中18只雌性小鼠,18只雄性小鼠。应用随机数字表进行随机分组,将雌雄小鼠各分为3组,分别为ERβR NAi组、空白对照组和阴性对照组。通过尾静脉注射转染ERβR NAi逆转录病毒载体和阴性对照组逆转录病毒载体,进行基因沉默的在体实验。各组小鼠均在相同条件下分笼喂养。分别于在体动物实验的当天、2周、4周和6周时测量各组小鼠身长、体重。于第6周时麻醉下处死小鼠,测量各组小鼠第一腰椎高度。应用SPSS16.0软件统计分析各组间差异的显著性。结果在雌性小鼠组中,ERβR NAi组小鼠身长在第2次和第3次测量时的增加值明显高于空白对照组和阴性对照组;ERβRNAi组小鼠体重增加值小于空白对照组和阴性对照组;ERβRNAi组小鼠身长/体重比在第2次和第3次测量时的增加值明显大于空白对照组和阴性对照组;差异有统计学意义(P<0.05);6周后测量ERβRNAi组小鼠第1腰椎高度明显大于阴性对照组和空白对照组(P<0.0)。而在雄性小鼠中,以上各检测指示均差异无统计学意义(P>0.05)。结论 ERβ基因沉默后,促进雌性小鼠脊柱生长。据此可以反向推断,雌激素与ERβ特异性结合,抑制雌性脊柱生长。
[Objective] To study the role of ERβ in the spinal vertebrae linear growth in adolescent. [Methods] 36 mice ,5-week old, the equivalent of prepubertal children, Of which 18 female mice, 18 male mice, were randomly assigned and divided into 3 groups using random number table, respectively, ERβRNAi group, blank control group and negative control group. Prepared gene silencing animal models by tail vein injection of transfected ERβRNAi retroviral vector and negative control retroviral vector. The mice were fed under the same conditions in different cages. After 6 weeks, The mice, equivalent to 18-year-old's human age or so, were anesthetized and lavaged, and the height of the first lumbar vertebral bodies were taken for testing. Mice animal model were measured the length (the length from tip to tail root, cm), weight (g), respectively, in 0 day, 2weeks, 4weeks and 6 weeks. SPSS16.0 software was used to analyze whether the differences was significantin. [Results] In female mice, the mice's length added value in the ERI3RNAi group at the 2nd and 3rd measurement were higher than the mice in control group and negative control group; The mice gained weight less in the ERI3RNAi group than in the other two groups; The ratio of height and weight in the ERβRNAi group at the 2nd and 3rd measurement were higher than in other groups, a significant difference, P 〈0.05. the height of the first lumbar vertebral body was significantly greater in the ERβRNAi group than in other groups, a significant difference, P 〈0.05. However, in male mice group, the above test results were not significantly different. [ Conclusion ] According to test results of the ER8 silence, we can reverselv infer that the combination of estrogen and ERβ-specific, directly and/or indirectly inhibit the linear growth of the spinal vertebrae.
