siRNA抑制甲胎蛋白表达对甲胎蛋白阳性胃癌细胞FU97增殖抑制及凋亡作用 被引量:2

摘要 目的观察siRNA抑制甲胎蛋白(AFP)表达对FU97细胞增殖及凋亡的影响。方法用抑制效率较高的siRNA-A1与siRNA-A3转染细胞,采用MTT法检测实验组载体对FU97细胞增殖抑制作用,DNA琼脂凝胶电泳法观察细胞凋亡后DNA切割片段。结果 siRNA-A1与siRNA-3质粒能显著性抑制细胞增殖,其抑制率随着作用时间延长而提高,抑制率最高可达到59.19%(P<0.05),siRNA抑制AFP基因表达后可诱导FU97细胞凋亡。结论 siRNA表达载体可有效抑制FU97细胞AFP基因表达,抑制胃癌细胞的增殖并诱导凋亡。 Objective To observe the effect of the expression of AFP repressed by siRNA on the proliferation and apoptosis of FU97 cell.Methods siRNA-A1 and siRNA-A3 of higher repressing rate were transfected to stomach cancer FU97 cells.The repression of the FU97 cells' proliferation was examined by MTT.The cell apoptosis was observated by DNA agar gel electrophoresis law.Result siRNA-A1 and siRNA-3 guilder shot can obviously repress cell proliferation,its represses rate was raising along with action time,the maximum of repressing rate can be 59.19%(P0.05).AFP gene repressed by siRNA can induce FU97 cells apoptosis.Conclusions siRNA can effectively repress AFP gene expression and proliferation of the FU97 cell,inducing apoptosis.
