摘要 目的探讨硫化氢对氧化高密度脂蛋白(ox-HDL)致人脐静脉内皮细胞(HUVECs)损伤的保护作用.方法体外培养HUVECs细胞,应用形态学观察法和噻唑兰(MTT)比色法检测细胞活性,试剂盒测定培养液中乳酸脱氢酶(LDH)及丙二醛(MDA)含量.结果ox-HDL组细胞损伤明显,经硫化氢低、中、高剂量组的干预,受损情况均有较明显改善,细胞存活率增加,且培养液中LDH及MDA含量降低,与对照组比较差异有显著性(P<0.05).结论硫化氢对ox-HDL所致内皮细胞损伤具有一定的保护作用,其机制可能与降低氧自由基抗氧化作用有关. Objective To investigate the protective effect of hydrogen sulfide on the injury of endothelial cell induced by ox-HDL. Methods HUVECs were incubated in vitro. The cells viability is measured by mor- phological observation and the MTT assay. The levels of LDH and MDA activity in culture medium were measured with corresponding kit. Results HUVECs were damaged obviously in ox-HDL group,hydrogen sulfide improved the condition of HUVEC significantly, increased the cell survival rate, decreased the lev- el of LDH and MDA(P〈0.05). Conclusion These results demonstrate that hydrogen sulfide can protect HUVECs from damage by ox-HDL,and it may be through decreasing oxygen free radical production and antioxidation.
