摘要
本工作以黄色节杆菌ATCC39106为出发菌株,通过诱变选育,获得突变株TS151,并研究了适用于13 C标记L-亮氨酸生产的发酵工艺、提纯工艺。结果显示,最佳发酵条件为:发酵温度28℃、220r/min摇床、培养时间92h。在此条件下,发酵产酸稳定在15g/L以上,提取收率稳定在70%。产品13 C标记L-亮氨酸丰度达到97.03%,纯度达98%以上。
With yellow Arthrobacter as starting strain and through mutation breeding, mutant strain TS151 is available to us . The dissertation focuses on the mutation breeding prescription ,fermentation condition and purification of L-leucine high-yielding strain TS151. The results show. With 92 h cultivation in 220 r/min rotation speed and 28℃ ,the acid production of high abundance is more than 15 g/L, extraction yield is about 70%. As products, the isotopic abundance of stable isotope labeled L-leucine is 97.03%, the purity is more than 98 %.
