宫颈鳞癌患者血清差异蛋白质与鳞状细胞癌抗原的关系 被引量:6

Relationship between differential proteins and squamous cell carcinoma antigen in patients with squamous carcinoma of the cervix

摘要 目的:研究宫颈鳞癌患者血清差异蛋白质与鳞状细胞癌抗原(squamous cell carcinoma antigen,SCCA)之间的关系,寻找早期诊断宫颈癌的生物学指标。方法:采用表面增强激光解吸离子化飞行时间质谱仪(SELDI-TOF-MS)和弱阳离子结合芯片(CM10),检测44例宫颈鳞状细胞癌及25例健康女性的血清蛋白质,将3组SCCA不同水平的宫颈癌患者分别与健康对照组进行对比,同时通过对比SCCA不同水平的宫颈癌患者,筛选出有分类意义的差异蛋白质。结果:(1)宫颈鳞癌SCCA正常患者与健康对照组比较,共有19种血清蛋白质质谱峰值差异具有显著性(P<0.001)。宫颈鳞癌SCCA升高患者(大于正常1倍)以及大于正常3倍组,分别与健康对照组比较,分别发现了26种和15种差异蛋白质(P<0.05)。其中有4种质荷比(M/Z)为4172、3397、2741、6086的差异蛋白质,重复出现于宫颈鳞癌SCCA正常或升高不同水平组与健康对照组的比较中。(2)在宫颈鳞癌患者中,SCCA小于正常3倍组与大于正常3倍组比较、SCCA小于正常4倍组与大于正常4倍组比较,分别发现了5种和20种差异蛋白质(P<0.05)。其中M/Z为8142、7765、3885、5632、9287的差异蛋白质,重复出现于上述两组比较中。结论:通过蛋白芯片-飞行质谱方法,能够更早的发现宫颈癌与正常人之间的血清差异蛋白质,这些差异蛋白质有潜力发展为宫颈癌早期诊断指标,有助于宫颈癌的筛查、区分早晚期宫颈癌以指导治疗,具有重要的临床应用价值。 Objective To investigate the relationship between differential proteins and squamous cell carcinoma antigen (SCCA) in patients with squamous carcinoma of the cervix. Methods The proteins in sera from 44 patients with squamous carcinoma of the cervix and 25 female normal controls were measured by surface enhanced laser desorption/ionization time of fight mass spectrometry (SELDI-TOF-MS) with weak action chips (CM10). 44 patients were divided into 3 groups according to the levels of SCCA, and the differential proteins were identified. Results As compared with those in control group, the peak mass spectrum of 19 kinds of serum proteins in patients with normal level of SCCA (P 0.001), 26 kinds in patients with 1-3 folds of normal level of SCCA and 15 kinds in patients with 3-4 folds of normal level of SCCA were different (P 0.05). Among which, there were 4 kinds of proteins with the values of 4172, 3397, 2741, and 6086 of M/Z could be found in all the patients. There were 5 kinds of different proteins between patients with 3-4 folds and 3 folds, 20 kinds between patients with 3-4 folds and 4 folds, among which, the proteins with the values of 8142, 7765, 3885, 5632, and 9287 of M/Z could be constantly found. Conclusion Differential proteins could be found in patients with squamous carcinoma of the cervix by SELDI-TOF-MS method.
