摘要
目的建立HPLC法测定肿瘤细胞中5-氟尿嘧啶的浓度。方法采用Diamonsil C-18柱(4.6 mm×250 mm,5μm),以甲醇-水(10∶90)为流动相,流速为1.0 ml.min-1,检测波长为265 nm,柱温25℃。结果线性范围为0.056 5~1.808 mg.L-1(r2=0.9998);回收率(n=5)大于96.6%;日间、日内精密度实验的RSD(n=5)均小于8.0%。结论该试验建立的方法简便、快捷、专属性强,可用于临床研究5-氟尿嘧啶的耐药机制,也可用于5-氟尿嘧啶的药代动力学的研究。
Objective To establish an HPLC mothod to determine 5-fluorouracil in tumor cells.Methods The column:Diamonsil C18column(4.6 mm×250 mm,5 μm)was adopted with the mobile phase of methanol-water(10∶ 90).The flow rate was 1.0 ml·min-1.The detection wavelength was at 265 nm,and the column temperature was 25℃.Results The calibration curves were linear between 0.0565 and 1.808 mg·L-1(r2=0.999 8).The recovery(n=5)was more than 96.6%.Intra-day RSD and inter-day RSD were both less than 8.0%(n=5).Conclusion The method is convenient and rapid with good specialization.It can be used for drug resistance mechanism study and pharmacokinetics study of 5-fluorouracil.
