摘要
目的检测人慢性粒细胞白血病K562细胞来源外泌体基质金属蛋白酶-9(matrix metalloproteinase-9,MMP9)和趋化因子受体-4(chemokine receptor 4,CXCR4)基因的表达,探讨K562细胞外泌体可能的功能。方法采用RT-PCR方法分别检测K562细胞和K562/Adr细胞(阿霉素耐药株)及两者分泌的外泌体MMP9、CXCR4基因的表达。结果在K562细胞和K562/Adr细胞中,均检测到MMP9和CXCR4基因的表达。而在K562细胞和K562/Adr细胞分泌的外泌体中,只检测到MMP9基因的表达。结论两种K562细胞都可以分泌一定量的外泌体;虽然两种K562细胞都表达MMP9、CXCR4基因,但外泌体只选择性携带和转运MMP9基因,推测其可能具有促血管新生功能。
Objective To detect the expression of MMP9 and CXCR4 in leukemic K562 cells-derived exosomes, and to explore the possible function clue of K562 cells-derlved exosomes. Methods Using reverse transcription polymerase chain reaction (RT-PCR) method to detect the expression of MMP9 and CXCR4 in K562 cells,K562/Adr cells, K562 cells-derived exosomes and K562/Adr cells-derived exosomes. Results MMP9 and CXCR4 were produced by K562 cells and K562/Adr ceils. But K562 cells-derived exosomes and K562/Adr cells-derived exosomes only expressed MMP9 gene. Conclusion CML cells secrete a cer- tain amount of exosomes. Although both K562 and K562/Adr cells express MMP9 and CXCR4, CML-derived exosomes express MMP9 selectively. So we infer that CML-derived exosomes may play a role in angiogenesis.
