抗酸-免疫组化双重染色在结核病组织病理学研究中的应用 被引量:2

摘要 目的:研究抗酸和免疫组织化学双重染色法在同一张切片上清晰显示结核分支杆菌与巨噬细胞的分布状态。方法:组织切片常规脱蜡、入水,其染色分别按抗酸-免疫组化-苏木素;免疫组化-抗酸-苏木素和免疫组化-苏木素-抗酸顺序进行。结果:组织切片按免疫组化-抗酸-苏木素顺序染色,背景清晰,反差明显,较另两种染色顺序显色效果好。结论:组织切片按首先免疫组织化学染色,在抗酸染色,最后苏木素染色的步骤染色效果最好。能在同一切片中清晰显示结核分枝杆菌与巨噬细胞的分布情况。 Objective: To find a proper order on Ziehl-Neelsen and immunohistochemical double staining to detect the distribution state of mycobacterium tuberculosis and macrophages in one section.Methods: The sections were dewaxed and rehydrated.The staining sequence in group A was Ziehl-Neelsen staining first,then immunohistochemical staining and finally for hematoxylin staining.In group B the staining sequence was immunohistochemical staining,Ziehl-Neelsen staining and then hematoxylin staining.The Ziehl-Neelsen and hematoxylin staining was in reverse order in group Cas compared with group B.Results: In group A,the result of Ziehl-Neelsen staining did not show.In group B,all of the staining results were existed.In group C,the result of hematoxylin staining did not show.Conclusion: First immunohistochemical staining,then Ziehl-Neelsen staining and finally hematoxylin staining is the best staining order in the experiment.Ziehl-Neelsen and immunohistochemical double staining could clearly show the distribution of Mycobacterium tuberculosis and macrophages in one section.
