摘要
探讨Zhangfei(ZF)在动情周期小鼠子宫和卵巢中的分布规律。通过阴道涂片方法确定昆明雌性小鼠的动情周期,利用免疫组化SP法检测了ZF在小鼠动情周期子宫和卵巢组织中的定位。ZF在各级卵泡的卵母细胞中均呈阳性分布,且主要定位于胞质中;次级卵泡和三级卵泡的颗粒细胞层中可见ZF的存在;成熟卵泡的颗粒细胞中ZF蛋白的阳性更加明显。ZF在各个动情时期的黄体中均有分布,而在凋亡的颗粒细胞和泡膜细胞中未见阳性产物分布。ZF在子宫中主要分布于内膜上皮细胞和腺上皮细胞,间质细胞与肌层中未见分布,ZF在间情期活动较强,发情期较弱。推测ZF可能与小鼠动情周期中卵泡发育和腺体分泌等有关。
To investigate the distribution of Zhangfei protein in mouse uterus and ovary during estrus cycle, the classification of estrus cycle stage of Kunming female mice was confirmed according to vagina smears, then protein localization of Zhangfei genes in the mouse uterus and ovary during estrus cycle was detected by immunohistochemical SP method. Positive staining for Zhangfei was present in granulosa cells from sec- ondary follicles and tertiary follicles, and the intensity was stronger in the granulosa cells of mature folli- cles compared with those of secondary and tertiary follicles. Zhangfei was also localized in the cytoplasm of the oocytes in all developmental stages of follicles. Zhangfei was also clearly found to be localized in the lu- teal cells of the corpus luteum in all stages of the estrus cycle, but absent in the apoptotic granulosa cells and theca cells. Zhangfei was specifically found in the luminal and glandular epithelial ceils of the uterus. However, no positive reaction was found in the endometrial stromal ceils or the cells of the myometrium. Zhangfei may play important roles in the regulation of follicular development and gland secretion functions during the estrus cycle.
