摘要
目的:通过脑脊液蛋白质组学实验技术体系,筛选并鉴定帕金森病患者脑脊液特异蛋白,初步探讨帕金森病的发病机制。方法:选自广州医学院第一附属医院神经内科与脑深部电刺激术(deep brainstimulation,DBS)治疗中心帕金森病患者和对照组脑脊液为研究对象,进行双向凝胶电泳,经银染、图像分析寻找差异蛋白质,并进行质谱分析鉴定。结果:鉴定得到4种差异蛋白质,分别是CENP-E、Tetranectin非钙离子C型凝集素样结构域、假想蛋白FLJ38159和KIAA1640。帕金森病组中KIAA1640蛋白含量占总蛋白含量的比例(0.09±0.05)%高于对照组(0.03±0.00)%,CENP-E(0.04±0.02)%、Tetranectin非钙离子C型凝集素样结构域(0.04±0.02)%、假想蛋白FLJ38159(0.11±0.06)%蛋白含量均低于对照组[(0.08±0.01)%,(0.07±0.02)%,(0.17±0.04)%],P<0.05。结论:本研究鉴定出4种差异蛋白,这些蛋白可能参与帕金森病的发病,但具体机制及其在帕金森病发病中的作用有待进一步研究。
Objective:Proteomics technology were applied to screen and identify disease-specific protein in eerebrospinal fluid (CSF) of patients with Parkinson's disease (PD), as an attempt to explore the pathogenesis of PD. Methods: The cerebrospinal fluid of PD patients and control CSF were collected at the Department of Neurology and Deep Brain Stimulation(DBS) Center of First Affiliated Hospital of Guangzhou Medical University. Two-dimensional gel electrophoresis with silver staining and subsequent image analysis was performed to screen for differential proteins which were then identify with mass spectrometry. Results:Four differential proteins were identified by mass spectrometry : CENP-E, calcium-free form of the C-type lectin-like domain of tetranectin, and two hypothetical proteins (FLJ38159 and KIAA1640). The PD group showed higher content of KIAA1640 protein [ (0.09 ±0.05 ) % vs ( 0. 03 ±0. 00) % ] as calculated by percentage of total proteins, lower contents of C ENP-E protein[ (0.04 ± 0.02)% vs (0.08 ±0.01 )% ], calcium-free form of the C-type lectin-like domain of tetranectin [ (0. 04 ±0.02)% vs (0. 07 ±0. 02 )% ] and hypothetical protein FLJ38159 [ (0.11 ±0. 06 )% vs (0.17 ±0.04) % ] compared with the control group ( all P 〈 0.05 ). Conclusion : The four types of differential proteins identified in this study may be involved in the pathogenesis of Parkinson's disease, although their specific mechanisms and roles remain to be further studied.
