摘要
以扇蕨品种铁扇无菌苗的叶片为外植体,研究了不同类型细胞分裂素对不定芽诱导、增殖和成苗的影响。结果显示:在附加2.0 mg/L 6-苄基氨基嘌呤(BAP)、激动素(KT)、苯基脲类细胞分裂素(CPPU)的改良MS培养基上,叶片均诱导产生不定芽,不定芽的诱导率分别为25.0%、40.6%和35.7%。转到添加0.5 mg/L CPPU的培养基上继代增殖,不定芽生长正常,芽繁殖系数达5.0以上;转到附加0.5 mg/L BAP、KT的培养基上继代增殖,不定芽生长受阻,转而诱导产生绿色小球,绿色小球的诱导率达95%以上,绿色小球的繁殖系数为5.0左右。不定芽和绿色小球在附加吲哚丁酸(IBA)0.2 mg/L的培养基上,成苗率达100%,发育成具有根、茎、叶的完整小植株。
The leaf of the sterile plantlet of Neocheiropteris palmatopedata(Baker) Christ was used as explant to study the effects of cytokinins of different types on its adventitious bud induction,proliferation and shooting.The results showed that in the modified MS medium with 2.0 mg/L 6-benzyl aminopurine(BAP),6-furifuryl aminopurine(KT) or N-(2-chloro-4-pyridyl)-N′-phenylurea(CPPU),the induction rate of adventitious buds was 25.0%,40.3% and 35.7% respectively.In the bud proliferation medium with 0.5 mg/L CPPU,buds grew healthily and the coefficient of bud propagation was above 5.0.In the bud proliferation medium with 0.5 mg/L BAP or KT,buds grew weakly,then were induced to green globular body(GGB).The induction rate of the GGB was 95%,and the propagation coefficient of the GGB was around 5.0.In the medium with 0.2 mg/L 4-(3-Indolyl) butyric acid(IBA),the adventitious buds and GGB developed into the intact regenerated plantlets with roots,stems and leaves,and the shooting rate was up to 100%.
