RP-HPLC法测定十味活血丸中延胡索乙素的含量

Determination of tetrahydropalmatine in Shiweihuoxue pill by RP-HPLC

目的建立反相高效液相色谱法测定＋味活血丸中延胡索乙素的含量。方法以十味活血丸为研究对象，采用DiamonsiIC18（4．6ram×200mm，5um）色谱柱，以乙腈．水．磷酸-三乙胺-（45：55：0．1：03，v／v／v／v）为流动相；流速：1．0mL·min-1；检测波长：280nm；柱温；35℃。结果延胡索乙素在1．04～10．4ug·mL-1（r=o．9998，n=6）浓度范围内与峰面积呈良好线性关系；平均加样回收率（n=9）为98。9％（RSD=1．1％）。结论本测定方法简便、快捷、准确，为测定十味活血丸中延胡索乙素的含量提供了准确可靠的分析方法。

OBJECTIVE To establish an RP＇HPIC method for the determination of tetrahydropalmatine inShiweihuoxue pill. METHODS The sample was analyzed in Diamonsil Ct8 column（4.6mm x 200mm, 5um）, with the mobile phase of acetonitrilewaterphosphoric acidtriethylamine （45 ： 55 ： 0.1 ： 03, v/v/v/v） at flow rate of 1.0mL· min-1. The UV detection wavelength was 280nm, and the column temperature was 35℃ . RESULTS The linear range was 1.04-- 10.4txg· ml.-1 （ r = 0. 999 8, n = 6） for tetrahydropalmatine. The average recovery of tetrahydropalmatine was 98.9 % （RSD = 1.1% ）. CONCLUSION The method is found to be simple, accurate and reliable for analysis of tetrahydropalmatine in Shiweihuoxue pill.
