Association between polymorphisms of epidermal growth factor+61 and susceptibility of hepatocellular carcinoma:A Meta-analysis

摘要 目的:系统评价表皮生长因子(EGF)+61基因多态性与肝细胞肝癌(HCC)易感性的关系。方法:计算机检索PubMed、MEDLINE和中国期刊全文数据库(CJFD)及万方数据库1980-2011年发表的有关EGF+61基因多态性与HCC易感性关系的相关文献。要求所纳入文献均为相关的病例对照研究,基因型在对照群体中的分布均符合H-W遗传平衡定律。采用RevMan 5.0进行Meta分析。结果:按照入选标准,有6篇文献包括7项病例对照研究纳入。Meta分析结果显示,与野生基因型AA比较,基因型AG并不增加罹患HCC风险〔OR=0.81,95%CI(0.70~0.95),P=0.008〕;基因型GG与HCC的遗传易感性相关〔OR=1.49,95%CI(1.28~1.74),P<0.01〕;与等位基因A比较,等位基因G增加罹患HCC的风险〔OR=1.38,95%CI(1.23~1.55),P<0.01〕。结论:EGF+61基因多态性与HCC易感性相关,等位基因G与基因型GG均增加罹患HCC的风险。 OBJECTIVE:To systemicly evaluate the relationship between polymorphisms of epidermal growth factor +61 and the susceptibility of hepatocellular carcinoma(HCC).METHODS: PubMed,MEDLINE,China Academic Journal(CJFD) and Wan fang database were searched for the literatures about the relationship between polymorphisms of EGF+61 and the susceptibility of HCC from 1980 to 2011.The literatures that case-control studies involving related subjects and genotype frequencies in control group consistent with Hardy-Weinberg equilibrium were included.The software RevMan 5.0 was used for Meta-analysis.RESULTS: According to inclusion criteria,there were six literatures included which included seven clinical trials.Meta-analysis showed,compared with the wild genotype AA,genotype AG did not increase the risk of suffering from HCC(OR=0.81,95%CI(0.70-0.95),P=0.008);Genotype GG was associated with the susceptibility of HCC(OR=1.49,95%CI(1.28-1.74),P0.01);Compared with the allele A,allele G increase the risk of suffering from hepatocellular carcinoma(OR=1.38,95%CI(1.23-1.55),P0.01).CONCLUSION: EGF+61 polymorphism is associated with the susceptibility of HCC,and allele G and genotype GG all increase the risk of suffering from HCC.
