肺癌患者外周血NKG2A和NKG2D表达及其临床意义的研究 被引量:7

Expression and clinical significance of receptor NKG2A and NKG2D in the peripheral blood of lung cancer patients

摘要 目的:通过研究肺癌患者外周血中自然杀伤细胞(NK)表面抑制性受体NKG2A和活化性受体NKG2D表达,分析探讨NKG2A与NKG2D表达失衡与肿瘤免疫逃逸的关系及其临床意义。方法:采用流式细胞术检测101例肺癌患者(肺癌组)和81例健康对照者(对照组)外周血中NK细胞含量及其表面受体NKG2A、NKG2D表达情况,分析NKG2A、NKG2D的表达与肺癌临床生物学特征之间的关系。结果:肺癌组外周血中NK细胞含量高于对照组,差异有统计学意义,t=2.25,P=0.025;肺癌组外周血中NKG2A表达高于对照组,差异有统计学意义,t=-3.66,P=0.001;肺癌组中NKG2A/NKG2D比值高于对照组,差异有统计学意义,t=-3.98,P=0.001;在肺癌组中随着TNM分期(F=4.99,P=0.009)和PS评分(t=-2.01,P=0.041)的增加,NKG2A表达也表现逐渐增加,差异有统计学意义,t=-2.01,P=0.041。结论:肺癌患者外周血中NK细胞表面受体NKG2A、NKG2D表达失衡,可能是肺癌发生免疫逃逸的机制之一。 OBJECTIVE:To observe the expression of inhibitory receptor NKG2A and activating receptor NKG2D on NK cells in the peripheral blood in lung cancer,investigate the possible relationship between the imbalance expression of NKG2A and NKG2D and tumor immune escape,and make a discussion on the clinical significance.METHODS: The content of NK cells and the expression of NKG2A and NKG2D on NK cells in the peripheral blood was detected in 101 patients with lung cancer and 81 healthy controls by flow cytometry analysis.The association of NKG2A and NKG2D levels with clinical features was analyzed.RESULTS: The content of NK cells in lung cancer group was significantly higher than that in healthy group,with statistically significant difference(t=2.25,P=0.025).The expression of NKG2A in lung cancer group was significantly higher than that in healthy group,with statistically significant difference(t=-3.66,P=0.001).The ratio of NKG2A and NKG2D in lung cancer group was significantly higher than that in healthy group,with statistically significant difference(t=-3.98,P=0.001).And with the increase of TNM stage(F=4.99,P=0.009) and PS grade(t=-2.01,P=0.041),the expression rate of NKG2A increased gradually,with statistically significant difference.CONCLUSION: The imbalance expression of NKG2A and NKG2D on NK cells in the peripheral blood in lung cancer may play important roles in lung cancer immunological escape.
