摘要
目的研究NF-κB和Caspase-3在肝细胞肝癌(HCC)中的表达及其意义。方法应用免疫组化技术对45例HCC组织及12例正常肝组织中NF-κB和Caspase-3抗原表达进行检测和分析。结果 NF-κB在正常肝组织、癌旁肝组织及HCC组织中表达率分别为16.67%(2/12)、28.89%(13/45)及62.22%(28/45)(P<0.05),同时NF-κB在HCC组织中的表达与肿瘤的分化程度及临床分期有关。Caspase-3在正常肝组织、癌旁肝组织及HCC组织中阳性率分别为83.33%(10/12)、68.89%(31/45)及42.22%(19/45)(P<0.05),Caspase-3在高、中、低分化HCC组织中的表达无显著性差异,但与肿瘤的分期有明显相关性。Caspase-3和NF-κB在HCC组织中表达呈负相关(P<0.05)。结论该实验结果提示NF-κB可能通过抑制Cas-pase-3的活性从而抑制细胞凋亡,在HCC的生长和进展过程中发挥重要作用。
[ Objective ] To study the expression and significance of NF-κB and Caspase-3 in hepatocellular carcinomas (HCC). [ Methods ] The immunohistochemical technique was used to detect the expression of NF-κB and Caspase-3 in 45 cases of HCC and surrounding liver tissue and 12 cases of normal liver tissue. [Results] Positive rate of NF-κB in HCC, surrounding liver tissue and normal liver tissue were 16.67% (2/12), 28.89% (13/ 45) and 62.22% (28/45)(P 〈0.05) respectively. The expression intensity of NF-KB was associated with the histological grade as well as clinical stage of HCC. Positive rate of Caspase-3 in HCC, surrounding liver tissue and normal liver tissue were 83.33% (10/12), 68.89% (31/45) and 42.22% (19/45)(P 〈0.05) respectively. The expression intensity of Caspase-3 was associated with the clinical stage, while there was no significant difference in expression of Caspase-3 between the highly differentiated, moderate differentiated and poorly-differentiated HCC. Negative cor- relation was observed between the expression of NF-κB and caspase- 3 in HCC. [ Conclusion ] It is suggested that NF-κB could prevent apoptosis through inhibiting activity of Caspase-3, which is important for the growth and development of HCC.
