硒拮抗砷抑制体外人胚肺组织中抗氧化酶活性的研究 被引量:8

摘要 目的观察硒和砷对组织中抗氧化酶活性的影响及硒对砷的拮抗作用。方法以体外培养人胚肺 组织为靶组织,使其暴露于不同浓度的As2O3和Na2SeO3。后检测其组织匀浆液中抗氧化酶GSH-Px(谷胱甘肽过 氧化物酶)、CAT(过氧化氢酶)、Mn-SOD(锰-超氧化物歧化酶)和总 SOD活性变化。结果 30、50、100μmol/L砷 使各酶活性显著降低;0.1和 1.0μmol/L硒使 GSH-Px及 CAT活性显著增高。预处理 0. 1和 1. 0μmol/L硒能明 显拮抗30、50μmol/L砷对4种抗氧化酶活性的抑制,但对100μmol/L砷引起的酶活性抑制作用不明显。10 μmol/L硒与各剂量砷呈抑制协同作用。结论适当剂量的硒能够拮抗一定浓度的砷对抗氧化酶活性的抑制,为 硒作为砷毒害的预防剂提供参考。 Objective Investigate the effect of selenite and arsenic on antioxidant enzymes activities in tis- sties and that of selenite antagonism toward arsenic. Methods Human fetal lung (HFL ) tissues were exposed to diverse concentrations of As2O3 and Na2SeO3. Then its activities of antioxidant enzymes GSH-Px,CAT,Mn-SOD and total SOD were measured. Results 30, 50, 100μmol/L arsenic significantly decreased the activities of 4 kinds of enzymes. On the contrary,GSH-Px and CAT activities of HFL tissues were markedly raised by supplementa- tion of 0.1, 1.0μmol/L Na2SeO3. The inhibition of 4 kinds of antioxidant enzymes activities caused by 30, 50μmol/L As2O3 could be significantly improved by pretreatment with 0.1, 1.0μmol/L Na2SeO3;Na2SeO3 had no effect on the inhibition of antioxidant enzymes activities caused by 100μmol/L As2O3; 10μmol/L Na2SeO3 preincubation with each concentration of As2O3 had a synergistic toxicity. Conclusions Appropriate dosage selenite can antagonize arsenic that induced inhibition of antioxidant enzymes activities. It will provide clues for selenium as a preventive agent to arsenic poison.
