摘要
目的探讨IL-1对血管形成促进因子与抑制因子间的动态平衡关系的影响,以进一步研究子宫腺肌病的发病机制。方法分离纯化培养子宫内膜间质细胞,并进行免疫组织化学鉴定。细胞传代后加入不同浓度IL-1,收集不同时间的培养液,采用ELISA法检测IL-8、VEGF、ENS的表达。结果子宫腺肌病的在位内膜间质细胞对IL-1作用的敏感性增高,产生较高水平的IL-8、VEGF(P<0.01)。一定浓度IL-1作用下,培养液中IL-8、VEGF的量随时间的延长而增加;不同浓度IL-1作用下IL-8、VEGF的量与IL-1剂量呈依赖关系。IL-1作用下的子宫腺肌病在位内膜培养液中ENS的表达与正常对照组无明显差异,且ENS的表达与IL-1作用的时间、IL-1的浓度改变无关(P>0.05)。结论子宫腺肌病在位内膜间质细胞在IL-1作用下IL-8、VEGF的表达均增强,而ENS的表达无明显改变。血管发生的平衡被破坏,致使内膜血管增生活跃,这可能是子宫腺肌病的发病机制之一。
【Objective】 To probe the effect of IL-1 on the dynamic equilibrium between VEGF and ENS and to study the paroxysm of endometioma at molecular level.【Method】 The interstitial cells were separated from endometrial and cultivated alone,and added different-potency IL-1 to the descents,collected different-period culture media,and to test the expression of IL-8,VEGF and ENS by means of ELISA.Meanwhile,immunohistochemistry tow-step procedure was used to test the cultivated cells and ascertain the purity of these cultivated cells.【Results】 In the case of endometioma,the sensibility of eutopic endometrial interstitial cells under the conditions of IL-1 increased;therefore high-tone IL-8 and VEGF came into being(P 0.01).Under the conditions of a certain-dense IL-1,the expression of IL-8 and VEGF increased with the time extension,which depended on time.The expressions of IL-8 and VEGF under the conditions of different-dense IL-1 depended on the dosage of IL-1.In the case of endometioma,the expression of ENS in the eutopic endometrial culture media,under the conditions of IL-1,had no obvious difference from the normal control group;moreover,the expression of ENS was not related to the change of time or dense of IL-1 at all.【Conclusion】 In the case of endometioma,the expression of IL-8 and VEGF ineutopic endometrial interstitial cells becomes strong under the conditions of IL-1;While the expression of ENS doesn't change obviously.It is the imbalance of angiogenesis.That leads to active growth of endometrial vascellum,which may be one paroxysm of the disease.
