摘要
根据NCBI上已知序列设计引物,利用RT-PCR技术从猪囊尾蚴中成功克隆了低分子量抗原基因Ag1V1,同源性和种系发育分析结果显示,河南分离株Ag1V1基因的核苷酸序列和预测氨基酸序列与日本分离株同源性为100%和98.9%,说明Ag1V1蛋白基因具有一定的保守性;通过生物信息学软件对其蛋白二级结构、疏水性及抗原表位等方面的预测,认为其有望成为诊断或免疫用抗原。
A pair of primers were designed based on Ag1V1 gene published in NCBI,Ag1V1 gene fragment was cloned successfully from Cysticercus cellulosae by RT-PCR.Sequence homology comparison results showed that Ag1V1 shared 100% similarity in nucleotide acid and 98.9% in amino acid with Japanese stain in GenBank.All of them were in the same evolutionary clade.It revealed that Ag1V1 protein was conservative in different regions.Secondary structure,hydrophobicity and antigenicity in Ag1V1 protein were predicted by bioinformatics tools.This information may be of help to the diagnosis or immunization of Cysticercus cellulosae in the future.
