摘要
目的:探讨参麦、葛根素注射液联合应用对大鼠肝缺血再灌注早期损伤的影响。方法:SD雄性大鼠50只,复制肝缺血再灌注损伤模型。随机分为:假手术组,模型组,参麦注射液干预组(5 mL/kg),葛根素注射液干预组(35m g/kg)及参麦、葛根素注射液联合应用干预组(3 mL/kg和25 m g/kg),每组10只。于再灌注后1 h检测肝组织丙二醛(M DA)含量及血清谷丙转氨酶(ALT)、谷草转氨酶(A ST)和乳酸脱氢酶(LDH)的含量;光镜下比较各组肝组织形态学的变化。结果:模型组血清ALT,A ST,LDH含量及肝组织M DA含量均明显高于假手术组(P<0.05);参麦、葛根素注射液干预组及二者联合应用干预组血清ALT,A ST,LDH及肝组织M DA含量均低于模型组(P<0.05);参麦、葛根素注射液联合应用组血清ALT,A ST,LDH及肝组织M DA含量的下降程度仅较单独使用二者时稍低,组间差异不明显,无统计学意义(P>0.05);光镜下参麦、葛根素注射液干预组及联合应用干预组的肝细胞损伤程度均较模型组明显减轻,但二者联合应用干预组的肝细胞损伤程度并未较单独使用二者时明显改善。结论:参麦、葛根素注射液对肝缺血再灌注损伤具有一定的保护作用,但二者联合应用不能进一步改善肝缺血再灌注所致的早期损伤。
Objective:To study the effect of shenmai and kakkonein injection combined application on hepatic is- chemia-reperfusion early injury in rats. Methods :Fifty SD male rats were randomly divided into the five groups with 10 animals each,They were sham operation group ,model group ,shenmai injection group(5 mL/kg),kakkonein injec- tion group(35 mg/kg),and shenmai and kakkonein injection combined application group (3 mL/kg and 25 mg/kg). The content of malondialdehyde (MDA) in hepatic tissues and serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) in serum were assayed after reperfusion for 1 hour. The in- jury circumstance of the liver tissue in each group was compared by optical microscope. Results :Hpatic tissue MDA and serum ALT,AST and LDH content in model group was higher than sham operation group(P〈0.05). Hpatic tis- sue MDA and serum ALT,AST and LDH content in shenmai group ,kakkonein group and combined application group deereased significantly compared with that in model group(P〈0. 05),and there was no difference between shenmai group,kakkonein group,and combined application group (P〉0. 05). The change could be observed by the optical mi- croscope in which liver cell injury of the combined application group was not slighter than that of the shenmai group and kakkonein group. Conclusion:Shenmai and kakkonein injection had a prominent protective effect on hepatic is- chemia reperfusion injury in rats,but shenmai and kakkonein injection combined application was not better than the single use of that.
