斑点热群立克次体中国分离株染色体DNA限制性核酸内切酶分析 被引量:5

摘要 应用限制性核酸内切酶Hind Ⅲ、BamHI、Pst Ⅰ及EcoRI,对本研究室从斑点热病人分离的立克次体An-84、Se-85、W-883株,从蜱分离的FT-84、MT-842株、从蜱卵分离的TO-85株及中国参考株JH-74蜱株与7株斑点热群立克次体国际标准株,进行了染色体DNA限制性核酸内切酶酶切片段电泳图谱分析。结果表明不同种的斑点热群立克次体国际标准株的DNA酶切图谱不相同,各具特征。7株中国分离株与国际标准株西伯利亚立克次体(Rickettsia sibirica)的DNA酶切图谱完全一致,株间未发现有差异。由此证明中国分离株均属西伯利亚立克次体,所致疾病为北亚蜱传斑点热(NorthAsia Tick-Borne Spotted Fever)。 Chromosome DNA from seven strains of spoted fever group (SFG) rickettsiae isolated in northern China and seven standard strains of SFG rickettsiae were analysed followed with restriction endonucle- ase Hind Ⅲ, Barn HI, EcoR I and Pst I, by electrop- horesis. The various kinds of standard strains of SFG rickettsiae had different DNA restriction pat- terns. The seven Chinese strains had identical DNA restriction patterns to those of Rickettsia sibirica (strains 232 and 246). These results demonstrate that seven Chinese strains of SFG ricketttsiae belong to Rickettsia sibirica. The results also sug- gest that this technique is useful in identification and classification of rickettsiae.
