二甲亚砜对脊髓损伤的抗自由基保护作用 被引量:1

摘要 目的探讨二甲亚砜(dimethyl sulfoxide,DMSO)对大鼠急性脊髓损伤后运动功能、自由基含量及组织病理学的影响。方法以改良Allen法制作脊髓损伤模型,将30只SD大鼠随机分为DMSO组、打击组和假手术组。术后分别观察24h、48 h、96 h的BBB评分;96 h后采血,检测血清标本中超氧化物歧化酶(SOD)和丙二醛(MDA)的含量;采血后取脊髓,苏木精-伊红(HE)染色观察损伤部位织病理学变化。结果大鼠后肢BBB评分术后96h DMSO组、打击组、假手术组BBB评分分别为(5.38±0.84)、(1.80±0.70)、(20.71±0.24)分,提示3个时相点均有所改善,且DMSO组均高于打击组,差异有统计学意义(P<0.05)。SOD检测分别为(149.08±16.02)、(131.36±19.66)、(158.57±14.69)U/mL,MDA检测分别为(4.85±0.33)、(6.36±0.49)、(3.683±0.25)nmol/mL,提示DMSO组自由基含量低于打击组,差异有统计学意义(P<0.05)。大鼠脊髓组织病理学切片观察显示,DMSO组炎症反应、结构破坏明显轻于打击组。结论 DMSO能明显促使脊髓损伤后的大鼠后肢的神经运动功能恢复,清除体内自由基,在一定程度上阻止脊髓继发性损伤的进一步发展。 Objective To investigate the effect of dimethyl sulfoxide(DMSO) on motor function,content of free radicals and histopathology after acute spinal cord injury in rats.Methods Animal model of acute spinal cord was induced through Allen method.30 SD rats were randomly divided into 3 groups: DMSO group,spinal cord injury group and sham-operation group.At last,the hindlimb motor function in the spinal cord were observed at the 24 h 48 h and 96 h after modeling.After 96 h of modeling,the content of MDA and activity of SOD in serum were measured for free radicals.The samples were taken from the spinal cord injury sites after haemospasia and tested by HE staining for histopathological.Results The hindlimb motor function of DMSO group and spinal cord injury group became better at the 24 h,48 h and 96 h,and there was obvious difference(P<0.05);Content of free radicals in DMSO group was much lower than that in spinal cord injury group after 96h of modeling(P<0.05);Histopathological observation showed that tissue damage to the structure of DMSO group significantly lighter than spinal cord injury group.Conclusion DMSO significantly clears away free radicals,accelerates the recovery of hindlimb motor function in rats after spinal cord injury and restraints secondary injury on the the neural structure.
