摘要
目的:探讨白介素21(IL-21)对细胞毒T淋巴细胞(CTL)杀伤非霍奇金淋巴瘤(NHL)的影响,阐述该影响是通过IFN-γ和TNF-α的高表达而实现。方法:分选树突状细胞(DCs),用NHL细胞株(Raji)冲击,诱导产生CTL;将此培养液分为对照组和实验组,实验组加入100ng/ml的IL-21。ELISA、RT-PCR检测IFN-γ和TNF-α水平。结果:IL-21作用组,IFN-γ水平:469.32±2.83pg/ml,TNF-α:82.52±4.72pg/ml,对照组,IFN-γ水平:27.63±2.76pg/ml,TNF-α:11.63±1.54pg/ml,两组差异显著(P<0.05)。结论:IL-21是通过IFN-γ和TNF-α的高表达而实现增强CTL杀伤NHL的作用,有重要的临床意义。
Objective:To investigate the effect of interleukin-21 on the anti-lymphoma activity of cytotoxic T lymphocytes induced by dendritic cells in vitro,and clarify the mechanism of CTLs against lymphoma is the expression of IFN-γ and TNF-α by CTLs.Methods:DCs was seperated from healthy human peripheral blood mononuclear cells.DC was attacked by Raji to induce lymphoma-specific CTL.Then the culture fluid was divided into control group and treatment group.The treatment group was given IL-21(100ng / ml).IFN-γ and TNF-α level in culture supernatant were determined by ELISA.Transcript expression of IFN-γ and TNF-α were detected by RT PCR.Results:After IL-21 acting on the CTL,IFN-γ level in the supernatant was 469.32 ± 2.83 pg / ml,and TNF-α was 82.52 ± 4.72pg / ml;no IL-21 acting group,IFN-γ was 27.63 ± 2.76pg / ml,TNF-α was 11.63 ± 1.54pg / ml,(P 0.05).RT-PCR results were the same.Conclusion:IL-21 can increase IFN-γ and TNF-α production in CTL to enhance it's anti-lymphoma effect.These results suggest that IL-21 have a potential applications in clinic.
