维甲酸诱导大肠癌CCL-187细胞成熟分化的研究

摘要 以全反式维生素甲酸 (RA)处理大肠癌细胞系CCL 187,观察RA对CCL 187细胞生长状态、细胞表面形态、超微结构、分化标志酶碱性磷酸酶活性以及内质网标志酶葡萄糖 6 磷酸酶活性的影响。结果发现 5 μMRA可明显抑制CCL 187细胞的生长 ,扫描电镜显示RA处理后的细胞表面较处理前规则 ,丝状伪足消失 ,出现钝形突起 ,超薄切片经透射电镜观察显示线粒体、内质网等细胞器增多 ,酶活性检测显示内质网标志酶葡萄糖 6 磷酸酶活性增强 ;分化标志酶 -碱性磷酸酶的活性较处理前明显升高 ,第 8天时较对照细胞高 2 3 4%。结果提示RA可诱导大肠癌细胞成熟分化。 Key Laboratory of Medical Cell Biology, Ministry of Public Heath of China,China Medical University. The changes of biological properties of cultured colorectal carcinoma(CCL 187) were studied during induced by all-trans retinoic acid (RA), such as cell growth、cell surface morphology、ultramicroscopic structure、activity of alkaline phosphatase and activity of endoplasmic marker glucose 6 phosphatase.The results showed that the growth of CCL 187 cells could be inhibited under 5μm RA significantly surface features of cells became regular. filopodia and surface blebs disappeared but blunt extensions appeared; the ultrathin sectioning displayed organelles such as mitochondria and endoplasmic reticulum were increased; activity of alkaline phosphatase was 234% higher than control cells after treated with 5μM retinoic acid,and activity of glucose 6 phosphatase was higher than untreated cells,too.The results indicated that RA could induced colorectal carcinoma differentiation.
