摘要
目的 环孢素亲合素 (cyclophilin ,CyP)是细胞内环孢霉素A(CsA)的受体 ,探讨CyP在肾病综合征患儿中基因表达及临床意义。方法 采用逆转录聚合酶链反应 (RT PCR)测定 2 8例肾病综合征患儿血白细胞中CyPmRNA的表达 ,并以正常儿童作对照。 结果 CyP碱基对 4 0 8bp与 β Actin条带 2 3 4bp可清晰区分。不同类型肾病综合征患者血白细胞中CyPmRNA表达的水平无差异 ,同时发现肾病综合征患儿急性期血白细胞中CyP基因表达水平明显高于恢复期与对照组。 结论 肾病综合征患儿急性期有高水平的CyPmRNA表达 ,在此时给予合适剂量环孢素亲合素的反应物(如CsA治疗 )可促进肾病综合征的缓解。
Objective To investigate the role and expression of cyclophilin(CyP)gene,receptor of cyclosporin A(CsA),in child nephrotic syndrome. Methods The authors studied the expression of CyP gene from white blood cells in children with nephrotic syndrome by reverse transcription polymerase chain reaction.Results It was shown that the base of CyP is helpful in differentiating 408bp from β Actin(234bp).The results also showed that the expression of CyP gene from white blood cells in children nephrotic syndrome in acute stage was significantly higher than those in restoring stage and those of controls.Conclusion These findings revealed a high level expression of CyP mRNA was observed in the acute stage of nephrotic syndrome,during which if timely CsA treatment at proper dosage is given,there might be a good prospect of remission of the disease.
