摘要 目的:探讨MRP基因、LRP基因和MDR1基因在胃癌中的表达及其与疾病发生发展相关性的研究.方法:应用实时荧光定量PCR(real-time fluorescence quantitative PCR,FQ-PCR)分别检测47例胃癌标本和17例正常胃组织对照标本的MRP基因、LRP基因和MDR1基因表达,分析其与病情发生发展以及转归的关系.结果:MRP基因、LRP基因和MDR1基因在胃癌组织中的表达均高于正常标本,MRP在早期胃癌中显著高于进展期胃癌(P<0.05),高、中分化腺癌显著高于低、未分化腺癌(P<0.05);LRP基因表达无淋巴结转移组显著高于淋巴结转移组(P<0.05).MRP在病情恶化患者中上调30%.结论:MRP基因、LRP基因和MDR1在胃癌组织中均有较高的表达,三者可能具有协同作用,检测3种基因的表达有利于制定更合理的治疗方案. AIM:To investigate the expression of the MRP,MDR1 and LRP genes in gastric cancer and to analyze their clinical significance.METHODS:The mRNA expression of the MRP,MDR1 and LRP genes in 47 gastric cancer specimens and 17 normal gastric mucosal specimens was detected by real-time fluorescence quantitative PCR (FQ-PCR).RESULTS:The expression of the MRP,MDR1 and LRP genes was significantly higher in gastric cancer than in normal gastric mucosal tissue.Positive expression of the MRP gene was correlated with tumor differentiation and stage,while LRP expression was correlated with lymph nodes metastasis.The expression level of the MRP gene increased by 30% in patients with progressing disease.CONCLUSION:The expression of MRP,LRP and MDR1 genes was significantly higher in gastric cancer than in normal gastric tissue and may be associated with multi-drug resistance in gastric cancer.
