摘要
利用 Pseudomonas Cepacia sp游离脂肪酶及其固相酶在非水相中催化外消旋体酯交换反应 ,对拟除虫菊酯手性农药中间体 (± ) - 4 -羟基 - 3-甲基 - 2 - (2 -丙烯基 ) - 2 -环戊烯 - 1-酮进行了拆分 ,考察了有机溶剂、水含量对酶活的影响。在实验确定的反应条件下 ,固相酶的酶活、热稳定性及操作稳定性优于游离酶 ,但选择性下降 ,目标产物的光学纯度分别为 73.5% ee和 94 .4 % ee。
Efficient kinetic resolution of a racemic ALON (±) 4 hydroxy 3 methyl 2 (2 propenyl) 2 cyclopenten 1 one,was achieved by Pseudomonas Cepacia sp Lipase and it's immobilized form in non aqueous organic media.The effects of organic solvent and water content on enzyme activity were studied. In experiment conditions,enzyme activity, thermal stability and operation stability of the immobilized lipase were more excellent than those of the free lipase.The optical purities of products for immobilized and free lipase were 73.5% and 94.4%, while the conversions were 56.2% and 35.6% respectively.
