摘要
[目的 ]研究甲基汞对急性分离的小脑神经细胞 [Ca2 + ] i 水平的影响及其机理。 [方法 ]利用 Fura- 2双波长荧光检测技术。 [结果 ] 1× 1 0 -7~ 5× 1 0 -6mol/ L甲基汞可使大鼠小脑神经细胞 [Ca2 + ] i 水平显著升高 (P<0 .0 1 ) ,且有良好的剂量 -效应关系。[结论 ]甲基汞升高神经细胞 [Ca2 -] i 的作用与胞外 Ca2 +大量内流和胞内储 Ca2 +释放有关 ,且以胞外 Ca2 +内流为主。胞外 Ca2 +内流与 NMDA受体门控 Ca2 +通道以及电压门控 Ca2 +通道有关 ,而与电压依赖性
Objective]To explore the effects and potential mechanisms of methylmercury on concentration of intracellular Ca 2+ in the acutely isolated cerebellar neurons [Methods]Fura 2 two wave length fluorophotometry was used [Results]1×10 -7 ~5×10 -6 mol/L methylmercury could increase the level of intracellular Ca 2+ of cerebellar neurons isolated from rats significantly (P<0 01) ,with a perfect dose response relationship [WT5”,6FZ][Conclusion]The increase action of methylmercury on intracellular Ca 2+ of neurons was associated with influx of Ca 2+ from outside of cells and release of stored Ca 2+ ,mainly with influx of Ca 2+ from outside of cells Furthermore,the latter was associated with NMDA receptor gated Ca 2+ channel and potential gated Ca 2+ channel,but not with potential dependent Na + channel
