摘要
以微生物絮凝剂产生菌EA为研究菌株进行种子培养和发酵,分析其絮凝活性的分布,然后对其发酵条件进行优化.选取不同的物质作为碳源、氮源,通过测量发酵产物的絮凝率来进行比较分析,对培养基初始pH、培养温度、接种量等主要发酵条件进行单因子实验和正交实验.结果表明:最有利的碳源氮源分别是葡萄糖和酵母膏;最有利的培养条件是初始pH值7.5、培养温度37℃、接种体积分数4%.经过优化培养之后发酵液对高岭土的絮凝率从71.7%提高到78.5%.
Microbial flocculant-producing bacteria EA was used as initial strain, the distribution of floccula-ting activity of which was analysed and then the fermentation condition was optimized. Different carbonsources and nitrogen sources were chosen to carry out comparison tests. The initial culture pH , cultivationtemperature, inoculation size were optimized through single factor experiment and orthogonal experiment. The results indicated that glucose and yeast extract were the best carbon source and nitrogen source, andthe best cultivation condition was initial pH of 7.5 ,cultivation temperature of 37℃ ,inoculation size of 4%.The flocculating activity could achieve 78.5 % under the best conditions.
