绝经后骨质疏松症肾阳虚证差异表达基因分析 被引量:16

摘要 目的通过基因表达谱的差比性分析,探讨绝经后骨质疏松症肾阳虚证相关基因的信息学特征。方法随机选择绝经后骨质疏松症受试者,分为肾阳虚证组3例,肾阴虚证组4例、非肾虚证组3例,并选择健康绝经后妇女3例设为正常对照组。用人全基因组表达谱芯片检测4组基因表达谱,肾阳虚证组分别与其他3组比较,筛选共同的差异表达基因,并对这些差异表达基因进行相关功能分析。结果肾阳虚证组与正常对照组、肾阴虚证组、非肾虚证组的差异表达基因分别为348,355,76条;肾阳虚证组与其他3组比较共同的差异表达基因有46条:与正常对照组相比,表达上调基因有4条,表达下调基因有42条。结论绝经后骨质疏松症肾阳虚证主要与免疫、细胞信号转导、钙离子活性和神经内分泌等方面相关。 Objective To explore the characteristics of genes expression profiles of postmenopausal osteoporosis patients with Kidney Yang deficiency through the variation analysis of gene expression spectrum.Methods Patients with postmenopausal osteoporosis were randomly chosen and divided into 3 groups,kidney yang deficiency group(n=3),kidney yin deficiency(n=4),and non-kidney deficiency(n=3).Three healthy postmenopausal women were selected in the control group.Human gene expression microarrays were applied to measure the gene expression spectrum of all patients in 4 groups.Patients in kidney yang deficiency group were compared with other three groups,respectively.The common variation genes were selected and the relative functional analysis was performed.Results The variation expression genes of patients in the kidney yang deficiency group were 348,355,and 76,compared with the control group,kidney yin deficiency group,and non-kidney deficiency group.There were 46 common variation expression genes of patients in the kidney yang deficiency group and other 3 groups.Compared with the control group,expression of 4 genes was up-regulated and expression of 42 genes was down-regulated.Conclusion The postmenopausal osteoporosis with Kidney Yang deficiency is mainly related with immunity,cellular signal transduction,Ca2+ activity,neuroendocrine,and so on.
