摘要
目的:优化虎杖中虎杖苷和大黄素提取工艺。方法:在单因素试验的基础上,以乙醇体积分数,提取时间和加液倍量为自变量,以虎杖苷和大黄素提取率为因变量,通过对自变量与因变量的完全二次响应曲面的回归拟合,用岭脊分析及效应面法优选虎杖最佳提取工艺,并预测最优工艺。结果:确定虎杖的最佳提取工艺为:以含醇量为59%的乙醇10倍量提取2次,每次提取1.1 h。结论:星点设计-效应面法可用于虎杖提取工艺优化,所建立的数学模型预测精确度高。
OBJECTIVE To optimize the process of extracting effective constituents from Giant Knotweed Rhizome. METHODS Based on the results of single factor experiment, taking ethanol concentration, extract time and solvents fold as the independent variables, while the extracting rates of polydatin and rheum emodin were taken as dependent variables, the central corn posite design and response surface methodology were used for fitting the relationship between dependent and independent varia hies,then predicted the optimal extraction process. RESULTS The optimum extraction processes were as follows: 10 fold 59% aqueous ethanol, extracting 1.1 hour for 2 times. CONCLUSION The observed values was close in agreement with the predicted values of the mathematic model, central composite design and response surface methodology is suitable for optimizing the extraction process.
